Abstract

Alginate lyaseAly448, apotential newmemberof thepolysaccharide lyase(PL)7 family,whichwas clonedand identified fromthemacroalgae-associatedbacterialmetagenomic library, showedbifunctionality.Themolecular docking results revealedthatAly448hastwocompletelydifferentbindingsitesforalginate(polyMG),poly-α-L-guluronicacid(polyG),andpoly-β D-mannuronic acid (polyM) substrates, respectively, whichmight be themolecular basis for the enzyme’s bifunctionality. TruncationalresultsconfirmedthatpredictedkeyresiduesaffectedthebifunctionalityofAly448,butdidnotwhollyexplain.Besides, Aly448presentedexcellentbiochemicalcharacteristics, suchashigherthermal stabilityandpHtolerance.DegradationofpolyMG, polyM,andpolyGsubstratesbyAly448producedtetrasaccharide(DP4),disaccharide(DP2),andgalactose(DP1),whichexhibited excellentantioxidantactivity.Thesefindingsprovidenovel insightsintothesubstraterecognitionmechanismofbifunctionalalginate lyasesandpaveanewpathfor theexploitationofnatural antioxidantagents.
