Abstract

Background and Objective: Interest in improving the agronomic traits of Leucaena tarramba has relatively increased to select productive varieties by plant breeding through gamma irradiation. Therefore, assessing the study of genetic diversity in Leucaena tarramba using RAPD markers is an important initial step to understand the genetic makeup of this species and its potential for improvement. Materials and Methods: In this study, RAPD markers was used to assess the genetic variability of some individuals based on the dose of gamma radiation treatment. By 10 primers used, four primers have successfully amplified the bands, consisting of OPA-05, OPA-09, OPD-03 and OPA-18 primers. Results: The informative band average of 3.25 NPB, 1.25 number of NMB and polymorphism of 21.32%. The genetic profiles such as, PIC, MI and EMR averaged 0.35, 17.60 and 44.50 per primer, respectively. Analysis of RAPD markers with dendrogram clustering revealed no significant genetic differences between Leucaena leucocephala and Leucaena tarramba. Irradiation dosages ranging from 250-750 Gy are closely linked, while a dose of 500 Gy shows the highest genetic distance. Conclusion: The informative band indicating a high level of effectiveness and reliability of the markers applied where 500 Gy is the most distant genetic relationship compared to other treatments. Furthermore, primer selection including OPA-05, OPA-09, OPD-03 and OPA-18, serves as a valuable information in biomolecular analysis of mutant Leucaena tarramba.
