Abstract

Satoimo (Colocasia esculenta (L) Scott var antiquorum) is one of the small corm taro.This commodity high
demand which is could not be fulfilled by the supplier has initiated some local Indonesian government to
develop satoimo as their superior export commodity. Tissue culture has become one of the propagation
techniques for satoimo development. The purpose of this research are to revealed the effect of 1,5
ppm 2iP, 25 ppm glutamin combined with three levels of GA3 (0,5; 1.0; 1,5 ppm) and 0,5 ppm NAA
combined with three levels of BAP (1; 2; 3 ppm) for satoimo in vitro multiplication. The result at 12
week after planting showed the highest shoot multiplication was in MS + 3 ppm BAP + 0,5 ppm BAP
(4,56 shoots). The higher BAP concentration up to 3 ppm BAP resulted in a higher shoot number
under the same NAA concentration (0,5 ppm). However the addition of 2iP, glutamin, and GA3 to MS
medium did not showed positive effect to satoimo in vitro shoot multiplication.
